FcγRs are members of the immunoglobulin superfamily that bind to the Fc portion of IgG. FcγRs are found on the surface of natural killer cells, monocytes, neutrophils and mast cells, playing a critical role in immunity28. In humans, there are three main types of Fcγ receptors: high‐affinity FcγRI and low‐affinity FcγRII and FcγRIII. These low‐affinity receptors are important in both promoting and regulating the immune and inflammatory response to immune complexes and are encoded by FCGR genes, located in a genetically complex cluster on the long arm of chromosome 1 (1q23)29. Furthermore, several recent studies indicate that FCGR genetic variants may be useful in predicting response to monoclonal antibodies in cancer and other inflammatory‐autoimmune diseases, such as rituximab, trastuzumab and cetuximab30,31. Genetic variation of FCGR2B is associated with response to immunoglobuline (IVIg) treatment in patients with Kawasaki disease32 and immune thrombocytopenia (ITP)33.
We have established a novel analysis able to genotypes Ile232Thr FCGR2B polymorphism. This method is based on real-time PCR. A genetic test for upfront treatment with IVIg led to faster recovery and less severe bleeding events.. User friendly and complete, the FCGR2B gene polymorphism Ile232Thr PCR real time detection kit is suitable for any laboratory.
